  • Abstrak : -Respon pertumbuhan lima aksesi talas beneng (Xanthosoma undipes k.koch) secara in vitro Growth response of five beneng taro accesses (Xanthosoma undipes k.koch) in vitro Adhilla Tsamrotul Fuadah, Prasetyorini, Surya Diantina* Program Studi Biologi, Fakultas Matematika dan Ilmu Pengetahuan Alam, Universias Pakuan, Jalan Pakuan, Tegallega, Kota Bogor, Indonesia – 16143 *Email: Adhillatsam@gmail.com INTISARI Penanaman secara kultur jaringan in vitro lima aksesi talas beneng (Xanthosoma undipes) dilakukan pada media dan unsur hara terbatas sehingga perlu penambahan ZPT (zat pengatur tumbuh) seperti kelompok sitokinin untuk mempercepat pertumbuhan tunas eksplan. Maka dari itu dalam penelitian ini ditambahkan ZPT kelompok sitokinin yaitu Benzyl Amino Purin (BAP) dan Thidiazuron (TDZ) pada media pertumbuhannya. Penelitian ini bertujuan untuk mengamati respon pertumbuhan lima aksesi talas beneng pada media pertumbuhan secara kultur jaringan in vitro dan membandingkan media perlakuan yang terbaik pada lima aksesi talas beneng. Penelitian ini telah dilaksanakan di BB BIOGEN. Waktu penelitian dilakukan selama tiga bulan yang dimulai dari bulan Oktober sampai Desember 2021. Rancangan penelitian yang digunakan adalah Rancangan Acak Lengkap (RAL) Faktorial terdiri dari 2 taraf perlakuan (A= Aksesi dan B= Media) dengan 5 kali ulangan yaitu A1 (talas juhut), A2 (talas juhut merah), A3 (talas kaduhejo), A4 (talas mandala), A5 (talas pasir peuteuy), dan B1 (MS), B2 (MS BAP 3 mg/L), B3 (MS BAP 5 mg/L), B4 (MS BAP 3 mg/L TDZ 0,1 mg/L AA 2 g/L), B5 (MS BAP 5 mg/L TDZ 0,1 mg/L AA 2 g/L). Dari hasil penelitian diperoleh bahwa respon pertumbuhan terbaik dari lima aksesi talas beneng yaitu A1 dan media perlakuan terbaik secara kultur jaringan in vitro yaitu B5. Pemberian MS BAP 5 mg/L TDZ 0,1 mg/L AA 2 g/L merupakan media perlakuan yang terbaik dan berpengaruh nyata terhadap parameter. Kata Kunci: Respon Pertumbuhan, Talas, dan Kultur Jaringan In Vitro. ABSTRACT Planting in vitro tissue culture of five accessions beneng taro (Xanthosoma undipes) was carried out on limited media and nutrients so it was necessary to Plant Growth Regulator (PGR) such as cytokinin groups to accelerate the growth of explant shoots. Therefore, in this study, cytokinin group PGRs, namely Benzyl Amino Purine (BAP) and Thidiazuron (TDZ) were added to the growth medium. This study aims to determine the growth response of five accessions beneng taro on growth media by in vitro tissue culture and compare the best treatment media on five accessions beneng taro. This research has been carried out at BB BIOGEN. The research was conducted for three months starting from October to December 2021. The research design used was a Factorial Completely Randomized Design (CRD) consisting of 2 levels treatment (A= Accession and B= Media) with 5 replications, namely A1 (juhut taro), A2 (juhut merah taro), A3 (kaduhejo taro), A4 (mandala taro), A5 (peuteuy taro), and B1 (MS), B2 (MS BAP 3 mg/L), B3 (MS BAP 5 mg/L), B4 (MS BAP 3 mg/L TDZ 0,1 mg/L AA 2 g/L), B5 (MS BAP 5 mg/L TDZ 0,1 mg/L AA 2 g/L). From the results of the study, it was found the best growth response of five accessions beneng taro was A1 and the best treatment medium in vitro tissue culture was B5. Giving MS BAP 5 mg/L TDZ 0,1 mg/L AA 2 g/L is the best treatment medium and has a significant effect on parameters. Keywords: Growth Response, Taro, and In Vitro Tissue Culture.
