Abstract
Forest betel (Piper aduncum L.) leaf is a plant that has the potential to be used as a traditional
medicine by Indonesian people. Forest betel leaf extract contains secondary metabolit compounds such
as alkaloids, tannins, saponins, flavonoids and terpenoids. This study aims to determine the
antimicrobial activity of forest betel leaf extract using multilevel Ultrasonic Assisted Extraction with
solvents of different polarities, namely n-hexane, ethyl acetate and 96% ethanol on the growth of
Streptococcus mutans bacteria and Candida albicans fungi. Antimicrobial activity was tested using the
paper disc diffusion method. The results showed that n-hexane, ethyl acetate and 96% ethanol extract
had antimicrobial activity with an inhibitory zone of 7.78 mm at a concentration of 30%, the 96%
ethanol extract was the most effective as an antibacterial in inhibiting S. mutans bacteria and the 96%
ethanol extract at a concentration of 45% is the most effective as an antifungal in inhibiting the fungus
C. albicans with an average inhibitory zone of 7.55 mm.
