ABSTRACT
Autoimmune or “lupus” is immune disorders caused by various factors, one of which is Streptococcus pyogenes bacteria. S. pyogenes bacteria activity can be suppressed using corticosteroid and antibiotic therapy, but has side effects. Tapak Dara (Catharanthus roseus `(L.) G. Don.) leaves known to have potency as an alternative antibacterial derived from secondary metabolites, namely 70 types of alkaloids in the form Triterpenoid Indol Alkaloids (TIAs), flavonoids, saponins, steroids, tannins, phenolics, antioxidants, and a little of essential oil. The purpose of this research are to determine the optimum concentration of 96% ethanolic and 96% methanolic extract from tapak dara leaves and the type of solvent extract which has the best antibacterial activity. Antibacterial potency test was carried out with dilution method to find KHM and diffusion to find DDH which in triplo at 96% ethanolic extract concentration of 45%, 55%, 65%, and 75%, while at methanolic 96% extract concentration of 15%, 25%, 35%, and 45%. The control was used DMSO 10% for negative control and 10 mg/L Amikacin antibiotic for positive control. Phytochemical tested with extract characterization and statistical test using Anova with α=0,05. Based are the results of the research, optimum concentration to inhibit S. pyogenes bacteria growth are concentration of 55% ethanolic extract with average diameter of inhibition area is equal to 9,20 ±0,05 mm and concentration of 45% methanolic extract with average diameter of inhibition area is equal to 19,583±0,803 mm. As for the solvent which has the best antibacterial activity is the methanolic solvent.
